The iso-IHC Detection Kits allow the detection of mouse antibodies on mouse tissues. The procedure suppresses the strong background caused by the cross-reactivity of the anti-mouse secondary and produces exceptionally high signal-to-noise ratios with strong staining intensities and no background. The method is based on the pre-labeling of mouse antibodies with an exclusive Biotin-labeling Reagent. This reagent biotinylates only mouse primary antibodies, even in the presence of contaminating proteins. Excess biotinylation reagent is blocked with the proprietary Mouse Block Reagent. The Biotinylated primary antibody mixture is added directly to the cell or tissues. The bound Biotinylated antibody is detected by the addition of a Streptavidin-conjugate that attaches strongly to the biotin residues on the primary antibody. If a Streptavidin-Enzyme conjugate was used, a chromogen/substrate is added and the resultant colored precipitate visualized by light microscopy.
